Principle of Nanopore Sequencing
Nanopore proteins are embedded in an insulating polymer membrane, with electrolyte solutions on both sides. Due to the negative charge of nucleic acids in solution, under an applied electric field, single-stranded DNA or RNA is driven from the negative electrode side (cis) toward the positive electrode side (trans) through the nanopore. Simultaneously, electrolyte ions flow through the nanopore, generating an ionic current across the membrane. As different nucleotides have distinct volumes and electrical properties, the ionic current fluctuates in a characteristic manner. By recording these current changes and interpreting the resulting signal patterns, sequencing is achieved.
